_All Fun and Games Until Someone Loses an Eye_, Christopher Brookmyre.

The movie The Long Kiss Goodnight takes a mythical uber-assassin, gives her an inconvenient case of amnesia, and reintroduces her a few years later as a normal person with a family. She begins to recover memories and skills...
Ludlum's _The Bourne Identity_ shows us the mythical uber-assassin with amnesia who immediately begins to recover skills, though not memories. The recent comedy Mr. and Mrs. Smith shows us two uber-assassins who keep their secret identities hidden from each other. No amnesia, just cover stories.

Brookmyre takes a Scottish grandmother and turns her into (what else?) an uber-assassin, laughing all the way. Somehow, it's all reasonably believable, too.

If I hadn't already been an unabashed fan of Brookmyre, this would have made me one. I imagine he has a great deal of fun writing.

Okay, so not *quite* like Mrs. Pollifax. :) (Have you read those?)

A grandmother at 42. Well.
